Using Fmoc-Asn(Trt)-OH and Fmoc-Gln(Trt)-OH in peptide synthesis prevents the possibility of the amide side chains of Asn and Gln from undergoing dehydration side reactions during activation, especially with carbodiimide reagents. Fmoc-Asn(Trt)-OH and Fmoc-Gln(Trt)-OH also have better solubility properties than Fmoc-Asn-OH or Fmoc-Gln-OH. Fmoc-Asn(Trt)-OH and Fmoc-Gln(Trt)-OH dissolve readily in all standard peptide synthesis reagents, while Fmoc-Asn-OH and Fmoc-Gln-OH are barely soluble in dimethylformamide (DMF).
